CTD profiles and underway seawater samples from INVT01_2024 (KSA324 assessment 2024)

This record describes the conductivity-temperature-depth (CTD) data and underway water sample data collected during the CSIRO Investigator transit voyage INVTO1_2024, between 9 March and 20 March 2024. The objective of the voyage was for a safe transit from Fremantle, Western Australia, to Hobart, Tasmania. This record was part of a piggyback project whereby undergraduate students attained at-sea training through the Institute of Marine and Antarctic Studies and the University of Tasmania. The project aim was for students to gain experience with oceanographic data collection, processing and analysis. The CTD data includes 7 CTD profiles that measured conductivity, temperature, pressure, dissolved oxygen, salinity, fluorescence, turbidity and photosynthetically active radiation (PAR). Each of these parameters were collected during each CTD profile. The CTD data also includes calculated measurements of ammonium, nitrate, nitrite, phosphate, silicate, as well as chlorophyll-a, from select rosette bottles. The underway water sample data includes 41 duplicate samples of calculated measurements of chlorophyll-a and associated calculated measurements of ammonium, nitrate, nitrite, phosphate and silicate nutrient concentrations. In both data files, ODV quality flags have been used to indicate data quality.

Statement
CTD Profiles: The Seabird 911plus CTD was deployed at seven stations along the voyage track. Two of the seven CTD profiles were to depths greater than 6000m. The CTD sensors were set up in duplicate to manage any redundancy. The seabird sensors measured dissolved oxygen, pH, turbidity, fluorescence, oil, PAR, nitrates, altimeter in real time. Once the CTD was returned aboard, seawater samples were collected from select rosettes for analysis of dissolved oxygen, salinity, nutrients, and chlorophyll-a. Underway water sampling: Chlorophyll-a and nutrient analysis was performed on 41 duplicate underway seawater samples collected from the underway seawater line. Each chlorophyll-a sample was collected in pre-rinsed 2L bottles and subsequently decanted into a 1L graduated cylinder. The samples were then filtered through a manifold with a funnel tower, using Whatman 25mm GF/F filters. Following filtration, filters were carefully removed using stainless steel forceps and transferred to glass scintillation vials. 10ml of cold 90% acetone was added to each vial and stored at -20 degrees Celsius. Non-acidified Chlorophyll a technique: Chlorophyll concentrations were determined using the non-acidification technique for extracted chlorophyll a (Turner Designs 2023). After 24 hours, samples were removed from the freezer, brought to room temperature by inversion and transferred to fluorometer vials. Three raw fluorescence values were taken using a Turner Trilogy fluorometer. Filters within the fluorometer are selective for chlorophyll-a. Prior to fluorescence measurement, three 90% acetone blanks were read, and occasional readings from a solid standard were taken to monitor instrument drift over time. Any acetone waste generated during the process was collected in a separate container for proper disposal. The average of three fluorescence readings was used to calculate chlorophyll a concentrations and all measurements were recorded along with corresponding latitude, longitude, and time in UTC. Nutrient sampling and analysis: Nutrient samples were collected from select CTD rosettes, as well as from underway seawater samples. The AA3 segmented flow analyser was used to determine ammonia, nitrate, nitrite, phosphate and silicate concentrations. When sampling for nutrients, vinyl gloves were worn to limit risk of contamination of samples. CTD nutrient samples were collected in Sarstedt Australia 10 mL polypropylene (PP) tubes with high density polyethylene (HDPE) screw caps (part number: 60.9921.819) (Rees et al 2019). Underway seawater samples were collected in scintillation vials. Sampling vials were rinsed three times prior to sample collection. After collection, samples were allowed to come to room temperature in the dark and measured within 2 hours to reduce any biological activity within the samples (Rees et al 2019). If analysis could not be completed within 2 hours, they were stored in the dark at 4°C and analyzed within 12 h.
