RNA-seq data from Ecklonia radiata exposed to single and repeated marine heatwaves

This dataset contains raw RNA sequencing reads from juvenile Ecklonia radiata (Laminariales) collected from Bruny Island, Tasmania, and used in a controlled laboratory heatwave experiment. Individuals were exposed to ambient, single, or repeated marine heatwave treatments to assess the effects of thermal stress and recovery. Total RNA was extracted from blade tissue and sequenced on an Illumina platform to generate paired-end reads. The data represent high-quality raw FASTQ files (n = 30 samples; 5 replicates per treatment per time point) suitable for transcriptomic analyses examining the molecular responses of E. radiata to marine heatwaves.

Statement
Juvenile Ecklonia radiata sporophytes were collected from Bruny Island, Tasmania, and acclimated in laboratory aquaria under controlled conditions. Individuals were exposed to three experimental temperature regimes simulating marine heatwave scenarios: no heatwave (15 °C throughout), a single 6-day heatwave at 20 °C, or two consecutive 6-day heatwaves at 20 °C separated by recovery periods at 15 °C. Samples for RNA sequencing were collected at two time points, during heatwave exposure and following recovery, from each treatment (n = 5 per treatment per time point). Blade tissue (approximately 50 mg) from each individual was preserved in RNA stabilization buffer and stored at –20 °C prior to extraction. Total RNA was extracted and purified before library preparation using the Illumina TruSeq Stranded mRNA Kit. Sequencing was performed by Macrogen Oceania (Sydney, Australia) on an Illumina NovaSeq X platform, generating 150 bp paired-end reads. This dataset contains the resulting raw FASTQ files for all samples.
